Pous Johannes XXI[1] is gebore as Pedro Julião (in Latyn Petrus Iulianus) of Pedro Hispano of Petrus Hispanus ongeveer in 1215 in Lissabon, Portugal. Hy is die seun van 'n dokter Julião Rebelo en sy vrou Teresa Gil. Tot in 2008 is hy die enigste pous van Portugese afkoms, hoewel Pous Damasus I as Portugees beskou kan word omdat hy gebore is in 'n gebied wat deesdae in Portugal is. Pous Paulus IV het wel 'n Portugese grootmoeder aan moederskant gehad. Pedro begin sy studies aan die biskoplike skool by die katedraal van Lissabon en later aan die Universiteit van Parys (ander bronne gee Montpellier). Hy studeer medisyne, teologie, logika, natuurkunde en metafisika. Tussen 1245 en 1250 het hy bekend geraak as Pedro Hispano omdat hy van Hispania (die Iberiese skiereiland) gekom het. Hy doseer geeeskunde aan die Universiteit van Sienna, waar hy sy boek "Summulae Logicales" (klein logiese somme) 'n teksboek met dertien dele oor die logika van Aristoteles wat nog vir 300 jaar gebruik sal word, skryf. By sy terugkeer na Portugal word hy raadgewer van koning Alfonso III en in 1273 aartsbiskop van Braga en kardinaal-biskop van Tusculum. Op 13 September 1276 volg Pedro Pous Adrianus V op as Pous Johannes XXI en regeer tot sy dood op 20 Mei 1277. Hy word opgevolg deur Pous Nicolaas III. Pous Johannes XXI[2] is op 20 September 1276 gekroon. Die pous het dadelik in die bul "Licet felicis recordationis"die streng reëls van die konsilie van Lyon wat bepaal het dat indien daar te lank gedraai word met die verkiesing van 'n pous, die kardinale se kos en drank verminder moes word, versag. Die pous se bewind is sterk beïnvloed deur kardinaal Giovanni Gaetano Orsini wat hom later sou opvolg as Pous Nicolaas III. Johannes het probeer om 'n nuwe kruistog te lanseer, versoening met die Oosterse Kerk te bewerkstellig en vrede tussen die Christen nasies te handhaaf. Hy het ook probeer om die Tartare te kersten, maar dit was vrugteloos.

Die pous het 'n nuwe vleuel aan sy paleis in Viterbo laat bou, die werk is sleg gedoen en terwyl die pous geslap het, het 'n gedeelte van die dak ingeval en die pous is ernstig beseer. Agt dae later op 20 Mei 1277 is hy aan sy verwondinge dood.
